In Word 2007, how do I get the definition of an acronym to show up when the
mouse is placed over the specific acronym?

What you are looking for is a feature used in PDFs & Web pages, not
supported in Word. The only thing that might give you something similar is
Comments, which you can find more on in Word Help.
